With AZ losing on Saturday, Ajax had the knowledge a win would move them further clear at the top. Hakim Ziyech returned to the starting eleven after making his comeback from injury in the cup midweek.
After 13 minutes, Donny van de Beek went down in the box and Dusan Tadic gave the hosts the lead from the penalty spot.
The hosts dominated the rest of the first-half but they could not find a second goal despite good opportunities. With one eye on the clash with Getafe next Thursday, Erik ten Hag decided to rest Ziyech at the break and bring in Daley Blind.
Seven minutes into the second half, Lassina Traore added the second goal with a close range finish. Klaas Jan Huntelaar then came off to add a third in the last minute.
Ajax move six points clear at the top and will now head to Spain for the Europa League clash with Getafe. RKC remains bottom.
