Ajax headed into the clash without Hakim Ziyech, David Neres and Daley Blind, while PSV were missing Steven Bergwijn, who left for Tottenham earlier in the week.
Carel Eiting started for Ajax and he set up Quincy Promes for the first big chance in the sixth minute. However, the winger was denied by Lars Unnerstall.
Ajax continued to dominate and Sergino Dest went close before Unnerstall denied Donny van de Beek. Eventually in the 35th minute, Promes opened the scoring with a calm finish from inside the box.
Ajax lost Promes to injury at the break with Lassina Traore coming in for him. The hosts continued to dominate the second half, but the game continued to be close.
PSV turned to top talent Noni Madueke late on and he came close to an equaliser but Lisandro Martinez intervened to earn his side the three points.
Ajax are now three points above AZ at the top of the Eredivisie while PSV sits 5th.
