Wolfsburg had the lead in the 15th minute through Weghorst, who then doubled the lead just before the hour mark. The Dutch striker now has five goals in four games, and 16 in the Bundesliga this season.
Kevin Mbabu made it 3-0 before Joao Victor added a fourth in the 69th minute. Rabbi Matondo pulled one back for Schalke but it was just a consolation.
The victory means Wolfsburg are sixth and can no longer fall out of the top seven, meaning a Europa League spot is definitely sealed with one game remaining.
